Teriparatide is the biologically active 1-34 N-terminal fragment of human parathyroid hormone (rhPTH(1-34); pharmaceutical brand Forteo/Forsteo; CAS 52232-67-4). Mechanism of Action

Teriparatide is an agonist at the parathyroid hormone-1 receptor (PTH1R), a class B GPCR expressed on osteoblasts and renal tubular cells. A key feature under study is that intermittent, pulsatile exposure is anabolic to bone — stimulating osteoblast activity and net bone formation — whereas continuously elevated PTH is catabolic. Signaling proceeds through Gs-adenylate-cyclase-cAMP/PKA and PLC pathways, with downstream effects on calcium/phosphate handling.
