PNC-27 (10mg) — Selective Anticancer Peptide | Oncology Research
PNC-27 is a 32-amino-acid chimeric peptide consisting of: (1) residues 12–26 of the p53 tumour suppressor protein’s MDM2-binding domain, fused to (2) a membrane-penetrating leader sequence from penetratin. The novel mechanism: HDM-2 (the human homologue of MDM2) is aberrantly expressed on the plasma membrane of many cancer cell types but not normal cells. PNC-27’s p53 domain binds membrane-associated HDM-2, causing the peptide to insert into the cancer cell membrane — creating transmembrane pores and direct membrane lysis. This mechanism is independent of p53 status, bcl-2, caspases, and standard apoptosis — making PNC-27 potentially effective against treatment-resistant cancers.
